利谷隆致雄性子代大鼠睾丸毒性的基因差异表达

摘要 目的探索利谷隆致雄性子代SD大鼠睾丸毒性的可能分子机制。方法实验分对照组和利谷隆组,分别于SD大鼠孕期12~17 d连续5 d灌服花生油和利谷隆,于SD大鼠孕期第20天解剖取胎鼠获得胎鼠每组40只,提取睾丸组织中RNA进行基因芯片分析。结果基因芯片分析仔鼠睾丸中有168个差异表达基因。结论利谷隆对SD雄性子代大鼠的生殖发育影响,可能是通过对睾丸发育相关基因及睾酮合成相关基因的异常表达改变的。 Objective To explore the testicular toxicity and its possible mechanism of linuron on F1 male rats.Methods Pregnant female rats were randomly divided into linuron group and control group,and exposed to linuron(dose,route) and peanut oil daily during the period of gestation days(GD) GD12-GD17,respectively,Anatomy the pregnant SD rats for 20 days to get fetuses(N = 40).The total RNA was extracted from the testide respectively and the whole genome microarray was hybridized to filter the differently expression genes in the testide from fetuses.Results Compared to control group,there were 168 differently expressing genes in the testesfrom the linuron-explosed group.Conclusion Linuron could induce testes development malformation in F1 male rats by altering the expression of genes related to testicle development and testosterone synthesis.
