摘要
以磺酰罗丹明染色法(SRB)法研究苦白蹄石油醚提取物(PEFO)体外对人肝癌细胞SMMC-7721、人胃癌细胞SGC-7901、人喉癌上皮细胞Hep-2三种人恶性肿瘤细胞增殖的影响。实验结果显示PEFO对三种肿瘤细胞都具有抑制作用,且呈剂量效应关系;其半数抑制浓度(IC50)分别为58.74、74.13、83.18μg/mL,对人肝癌细胞SMMC-7721表现出较强的抑制作用。通过倒置相差显微镜、透射电镜观察到SMMC-7721细胞经PEFO处理后,其形态和细胞超微结构发生较大改变,细胞出现核皱缩、贴壁性差、细胞裂解等现象;且细胞线粒体发生肿胀,胞质内出现大量空泡化结构。流式细胞术检测结果发现SMMC-7721细胞经PEFO处理后,S期细胞减少,G0/G1期细胞增多,出现了G0/G1期阻滞。研究结果显示PEFO在体外能够抑制肿瘤细胞的增殖,并初步推断有可能是通过抑制肿瘤细胞有氧代谢和干预细胞周期来实现抗肿瘤作用的。
The sulforhodamine B (SRB) assay was employed to evaluate the anti-tumor effect of petroleum ether extracts from Fomitopsis officinalis Ogill.:Fr.) Bond 0aEFO) on SMCC- 7721,SGC-7901,and Hep-2 cell lines in vitro. The results showed PEFO inhibited the growth of three kinds of cells in dose-dependent manner with the ICs0 values on SMCC-7721,SGC-7901,and Hep-2 cells being of 58.74,74.13 and 83.18 ktg/mL, respectively. The SMCC-7721 cells exposed to PEFO obviously changed cell morphology and cellular ultmstructure. The SMCC-7721 cell presented wrinkled nuclei, poor cell adherent growth and cell lysis, while mitochondria swell and a large number of vacuoles in cytoplasm were observed by transmission electron microscope. The flow cytometry results revealed that SMCC-7721cell decreased in S-phase and increased in Go/G^-phase, which arrested in Go/G1 phase. The study demonstrated that PEFO could inhibit cell proliferation in vitro, and the anti-tumor activity of PEPO might be achieved by inhibition of tumor cellular aerobic metabolism and intervention cell cycle.
