摘要
本文根据A.Klug,R.Henderson和L.A.Amos等人提出的付利叶空间的三维重构算法,对猪心肌Z盘的电镜图象进行了三维重构,系统地完成了付利叶空间三维重构的全部过程,得到了猪心肌Z盘的三维结构,从我们的结果可以看出,猪心肌Z盘的结构是近似的二维晶体,晶胞近似呈正方形,晶胞的大小约在220A左右。在每个晶胞中,质量分布由集中于晶胞中心而逐步扩散,最后分别集中到晶胞的四角上,我们所得的三维重构结果的分辨率约为70-80A,这些成果作为研究大型脊椎动物心肌Z盘结构的初次尝试,在生物、医学上是有意义的。我们所用的程序和计算方法有较大的普适性,可以作为研究生物大分子结构的基础。
Based on A. Klug, L. A. Amos and R. Henderson's fourier transform three-dimensional (3D)reconstruction method, the 3D reconstructed structure of the Z-disk of pig cardiac muscle from its electron micrographs has been obtained. Both theoretical and experimental aspects of 3D reconstruction method have been discussed in detail. The reconstructed result have also been processed and analysed by computer.Accordigng to the characteristics of pig's Z-disk and the analytical instrument conditions, the measure tech-niques and the image processing methods have carefully been selected and thoroughly been considered. A series of modifications and supplements have been done to the 3D reconstruction computer programs.The results of 3D reconstruction show that the Z-disk of pig cardiac muscle is a square-like lattice and the unit cell is about 220 A large. In each unit cell the mass distribution spreads from the center and then converges gradually to the four corners of the unit cell. The resolution of the 3D reconstruction result is about 70 A-80 A. although the Z-disk specimen wer used is not a very good two-dimensional crystal that limits the resolution of our 3D reconstruction result, the 3D structure of the Z-disk of pig cardiac muscle we obtained is of signifi-
cance in both biology and medical science.
