摘要
目的观察消毒愈肌膏对大鼠糖尿病足溃疡组织骨形态发生蛋白-9(bone morphogenetic protein-9,BMP-9)和转化生长因子β1(transforming growth factor-β1,TGF-β1)表达的影响。方法选用链脲佐菌-素腹腔注射与薄铁片固定法制备模型,按随机数字表法分为中药组、模型组、BMP-9对照组和TGF-β1对照组,每组20只,并将大鼠足部溃疡处分别外敷消毒愈肌膏和无菌盐水,腹腔注射外源性BMP-9、TGF-β1,30天后取四组溃疡组织标本,采用miRCU-RYTM芯片检测各组基因并分析基因表达。结果 BMP-9对照组和中药组中BMP-9基因表达明显增加,与模型组相比有显著差异(P≤0.01),BMP-9对照组与中药组BMP-9基因相比有显著差异(P≤0.05);TGF-β1对照组和中药组TGF-β1基因表达明显增高,与模型组相比有显著差异(P≤0.01),TGF-β1对照组与中药组相比,TGF-β1基因表达无明显差异(P≥0.05),中药组中BMP-9和TGF-β1表达变化呈显著正相关。结论消毒愈肌膏能显著调控BMP-9、TGF-β1表达水平,间接刺激组织细胞的分裂增殖,以利于创伤修复。
Objective To investigate the effects of Xiaodu Yuji Paste on treatment of diabetic feet from the level of genome. Methods 80 rats of diabetic foot ulcer were randomly divided into four groups (20 each) to receive traditional Chinese medicine, intraperitoneal injection of exogenous BMP-9, TGF-β1 and saline for 30 days. Total RNA was extracted from the tissue collected near the wound, and the expression profile of gens demonstrated using the miRCU-RYTM for rats. Results The levels of serum BMP-9 in BMP-9 group and TCM group were significantly higher than those in model group . The levels of serum BMP-9 in TCM group were significantly higher than those in BMP-9 group. The levels of serum TGF-β1 in TGF-β1 group and TCM group were significantly higher than those in model group. There are not difference in the levels of serum TGF-β1 in TGF-β1 group and TCM group. The levels of serum BMP-9 and TGF- β1are significantly positively related to change in TCM group. Conclusion These findings suggest that the Xiaodu Yuji Paste can regulate the differential expression of some genes. Indirect stimulation of cell prolif- eration, in order to facilitate wound healing.
