“益气调血,扶本培元”针法对SAMP8痴呆鼠大脑皮层脂筏募集细胞骨架相关蛋白的影响 被引量:4

摘要 目的研究针刺治疗阿尔茨海默病(Alzheimer's disease,AD)细胞骨架相关的细胞生物学机制。方法以SAMP8鼠为阿尔茨海默病动物模型,用Morris水迷宫检测"益气调血,扶本培元"针法对SAMP8鼠痴呆状况的改善效果。采用液质联用蛋白质组学方法检测该针法对SAMP8痴呆鼠大脑皮层脂筏募集细胞骨架相关蛋白种类的影响。结果与非穴组相比,针刺穴位不仅明显改善SAMP8痴呆鼠的认知能力,同时还能使SAMP8痴呆鼠皮层脂筏蛋白数量明显增多,有163种新蛋白被募集到脂筏中,其中细胞骨架蛋白和小G蛋白分子的增加非常显著,并且呈现出同步增多的特点;募集的小G蛋白分子主要是Rho家族和Ras家族成员,骨架蛋白主要是微管组装相关蛋白以及微丝动力蛋白家族成员。结论细胞骨架重组和依赖细胞骨架的胞内物质运输环节是针刺治疗阿尔茨海默病的效应靶点。 Objective To investigate the cytoskeleton-related cytobiological mechanism of acupuncture treatment for Alzheimer's disease (AD). Methods The SAMP8 mouse was used as a animal model of Alzheimer's disease. The improving effect of qi-replenishing, blood-regulating, body resistance-increasing and kidney-reinforcing acupuncture on dementia in SAMP8 mice was evaluated using a Morris water maze. The effect of this acupuncture method on cerebrocortical lipid raft recruitment of cytoskeleton-associated protein in SAMP8 mice was assessed by liquid chromatography-mass spectrometry-based proteomics. Results Compared with the non-acupoint group, acupuncture at points not only markedly improved cognitive ability but also significantly increased cortical lipid raft proteins in SAMP8 mice. One hundred and sixty-three kinds of new proteins were recruited to lipid rafts. Of them, cytoskeletal proteins and small molecular G-proteins increased very significantly, which was characterized by simultaneous increases. Recruited small molecular G-proteins were mainly the members of Rho and Ras families. Cytoskeletal proteins were mainly microtubule associated proteins and the members of microfilament motor protein family. Conclusion Cytoskeletal reassembly and cytoskeleton-dependent intracellular transport links are the effect targets for acupuncture in the treatment of Alzheimer's disease.
