摘要
为了获得酶溶性罗非鱼鳞胶原蛋白的最佳提取工艺,采用响应面法进行了提取工艺的优化。在单因素试验结果的基础上,根据中心复合设计试验设计原理,以胶原蛋白提取率为优化目标,建立了提取率与酶与底物比、时间、温度、液料比的数学模型。结果表明,酶与底物比、时间、温度对提取率的影响显著,最佳提取工艺为:酶与底物比3.4%、时间4.0 h、温度59℃、液料比35∶1,此条件下的提取率为13.12%,与预测值无显著性差异。试验所得到的数学模型为酶溶性罗非鱼鳞胶原蛋白的提取提供了依据。
In order to obtain the best extraction process for enzyme soluble tilapia scale collagen, response surface methodology was used to optimize the extraction process. On the basis of single factor test results and according to central composite design experimental design principles, a mathematical model was established to describe extraction yield as a function of enzyme substrate ratio, time, temperature, and liquid-solid ratio. The results showed that enzyme substrate ratio, time and temperature had significantly effect on the extraction yield, and the optimal extraction process obtained was as follows: enzyme substrate ratio 3.4%, time 4.0h, temperature 59℃, and liquid-solid ratio 35:1. Under these conditions, the extraction yield was 13.12%, which has no significant difference between the experimental and predicted values. The mathematical model obtained will provide a basis for the extraction of enzyme soluble tilapia scale collagen.
