裂叶马兰无性系建立的研究

摘要 为保护野生资源、满足栽培对种苗的需求,以裂叶马兰的嫩茎为外植体,进行愈伤组织诱导与分化、不定芽生根、试管苗生根继代增殖的研究,以及试管苗移栽和定植的研究,建立起裂叶马兰的无性系。结果表明:MS+蔗糖35 g·L-1+6-BA0.5mg·L-1+2,4-D2.5 mg·L-1是嫩茎愈伤组织诱导培养和继代增殖培养的适宜培养基;MS+蔗糖30 g·L-1+AgN031.0 mg·L-1+GA31.0mg·L-1+ZT1.6 mg·L-1+NAA0.1 mg·L-1是愈伤组织分化培养的适宜培养基;1/2MS+蔗糖20 g·L-1+NAA 0.6 mg·L-1是不定芽生根培养和试管苗生根继代增殖培养的适宜培养基;试管苗移栽成活率为95.9%;定植成活率98.0%。定植成活的试管苗保持了野生裂叶马兰的所有植物学性状。 In order to preserve the wild resources and meet the cultivated needs of people, with the method of tissue culture, the tender stems of Kalimeris indica (L.)Seh.-Bip were used as materials to do the research on callus inducement and differentiation, rooting of adventitious buds, rooting multiplication of tube seedlings, transplantation of tube seedlings and stable planting. The results demonstrated that MS+ sucrose 35 g·L-1 +6-BA0.5mg·L-1+2,4-D2.5mg·L-1 was the ideal medium for callus inducement and callus multiplication; MS+ sucrose 30 g·L-1+AgNO3 1.0mg·L-1+GA3 1.0 mg·L-1+ZT 1.6mg·L-1+NAA 0.1mg·L-1 was the optimum medium for callus differentiation cultivation; The ideal medium for rooting of adventitious buds and rooting multiplication of tube seedlings was 1/2MS+sucrose 20 g·L-1+NAA0.6 mg·L-1. The transplanting survival rate of tube seedlings was 95.9%. The planting survival rate of tube seedlings was 98.0%.The tube seedlings stable planted maintained all botany characteristics of the wild Kalimeris indica(L.) Sch.-Bip.
