铝试剂共振散射法测定血红蛋白

摘要 文章使用共振散射法,选择铝试剂,提出了一种测定血红蛋白的方法.体系使用pH为4.88的醋酸钠-醋酸(NaAc-HAc)缓冲溶液;铝试剂与血红蛋白发生反应,使体系的共振散射强度增强.最大散射波长位于438 nm处,在最佳条件下,测定血红蛋白的线性范围是0.048~9.60μ g/mL,线性回归方程:△I=18.19*Cm-3.956,相关系数R=0.9960,检出限为0.031 μg/mL.将方法用于人尿液中血红蛋白含量的测定。 A novel method which could be used to analyze the concentration of hemoglobin(Hb) was put forward. And the organic dye: aluminon, was selected in the method. The NaAc-HAc buffer solution of pH4.88 was used in the reaction-system. The Resonance Rayleigh scattering(RRS) signal could be increased obviously after adding aluminon in Hb solution. The maximum wavelength located at 438 nm. Under the optimized conditions, it was proportional to the concentration of lib in the linear range from 0.048 μg/mL to 9.60μ g/mL. Linear equation was: △I=18.19*CHb-3.956, correlation coefficient(r) was 0.9960. The detection limit(DL) was 0.031 μg/mL. The new method was built and used to analyze the healthy human urine samples with satisfactory results.
