摘要
目的:建立用苯酚-硫酸法测定黄精总多糖含量的方法。方法:以葡萄糖为对照品,在490nm处以浓硫酸的用量、反应温度和反应时间为考察因素,优选最佳显色条件。结果:苯酚-硫酸法测定多糖的最佳显色条件为加浓硫酸7.0mL、反应温度50℃和反应时间20min,葡萄糖浓度在12.16~60.80μg·mL。范围内与吸光度呈良好的线性关g(r=0.9994)。结论:用该法测定黄精总多糖含量,快速准确,稳定可靠,可作为黄精总多糖含量测定方法。
Objective: To establish the content determination method of total polysacchafides in polygonatum. Methods: Total polysaccharides were assayed by phenol-sulfuric acid method, referred to the grape sugar, and optimized the best color conditions, which regarded the dosage of sulfuric acid, reaction temperature, and reaction time as observation factors. Results: The best color conditions of polysaccharides which were determined by phenol-sulfuric acid method were sulfuric acid 7.0 mL, reaction temperature of 50℃ and reaction-time 20 min; the quality of glucose at the range of 12.16~60.80μg·mL had a good linear(r = 0.9994); Conclusion: Phenol-sulfuric acid method used to determinate the content of total polysaccharides in polygonatum is fast, accurate, stable and reliable, which can be regarded as content determination method for the total polysaccharide.
