胶体金免疫层析法快速检测水产品中甲基睾丸酮残留

摘要 研究基于免疫竞争法快速检测水产品中甲基睾丸酮残留含量的胶体金免疫层析试纸的制备方法。将抗甲基睾丸酮单克隆抗体-胶体金复合物包被在胶体金结合垫上,并将人工合成的甲基睾丸酮抗原包被在硝酸纤维素薄膜表面作为检测线(T线),其与待测样品中甲基睾丸酮竞争结合胶体金标记的甲基睾丸酮单克隆抗体,并能以颜色直观显示检测的定性结果。该试纸条可以在15 min内完成对水产品样本的半定量检测,灵敏度最低值可达到10 ng/g。试纸条具有较高的灵敏度及特异性,操作便捷,稳定可靠,可作为甲基睾丸酮残留现场监控的有效筛检手段。 A rapid, simple colloidal gold immunochromatographic assay for methyltestosterone (MT) residues was es- tablished based on immune competition principle. Nanocolloidal gold particles were prepared, labeled with an anti - methyhestosterone monoelonal antibody, and nitrocellulose (NC) membrane strip was separately coated with synthetic MT antigen as the test line(T) , which can combine with MT monoclonal antibody labeled by colloidal gold competi- tively in the samples. The method was used to detect MT residues in aquatic products, and semi - quantitative detec- tion results can be obtained within 15 minutes, and the lowest sensitivity can reach 10 ng/g. It was shown that the concentration level was positively related to the color change. As a result, the strip has high sensitivity and specifici- ty, being convenient, stable and reliable, and can be used as an effective screening method for MT monitoring.
