摘要
为建立稳定的草莓不定芽再生体系并为基因工程遗传改良奠定基础,以红颊和章姬无菌系叶片为外植体,研究了不同培养基、不同暗处理时间及不同叶龄对不定芽再生的影响。结果表明:2个品种在MS+B5+TDZ(3.0mg/L)+NAA(0.1mg/L)培养基上不定芽的再生率最高,单叶平均再生芽数最多;暗处理2周时间可有效促进不定芽的再生;叶片再生的最佳叶龄为40d,红颊和章姬的再生率分别达94.17%和95.83%。
In order to build a stable adventitious bud regeneration system of strawberry and lay a foundation for genetic improvement of genetic engineering, taking sterile system leaves of Hongjia and Zhangji as explants, the effects of culture medium, dark-processing time and leaf age on adventitious bud regeneration were studied. The results showed that, medium of MS+ B5 q- TDZ(3.0) +NAA(0.1 ) got the highest regeneration rate and the average number of regeneration buds was the most. Dark culture for 14 days could effectively promote the regeneration of adventitious buds. The best leaf age of adventitious bud regeneration was 40 days, and the regeneration rate of Hongia and Zhangii was 94. 17% and 95. 83% respectively.
