摘要
为研究谢瓦氏曲霉间型变种中nsdD基因的结构及其与有性产孢的关系,根据同源克隆及染色体步移技术克隆nsdD的全基因序列,并进行生物信息学分析。结果表明:该序列基因全长为1 773bp,开放阅读框长度为1 293bp,编码430个氨基酸,分子质量为46.659 4KD,理论等电点为9.08。序列同源性分析该基因与土曲霉(Aspergillus terreus)nsdD基因相似度最高为69%,含有1个GATA型锌指保守结构域,与其他曲霉的nsdD蛋白保守域相符。
To explore the relationship between nsdD gene structure and sexual reproduction, the complete DNA of nsdD gene was cloned by homology cloning and genome walking technology and was analyzed by bioinformatics. The results showed that full-length genomic sequence of nsdD is 1 773 bp, the gene contains a open reading frame of 1 293 bp and encodes 430 amino acid residues. The estimated molecular weight and isoelectric point of the putative protein respectively are 46. 659 4 kD and 9. 08. Analysis of the nucleotide sequence indicated that it had the highest 69% similarity with A. terreus nsdD gene and contained a GATA type zinc finger DNA-binding domain, the domains were conserved in other Asl)erillus.
