摘要
采用CTAB改良法提取葡萄属 7种植物的基因组DNA ,对其完整性进行了测定 ,并以此为模板进行RAPD反应 .对RAPD反应体系的 5个主要影响因子进行系统的初选与复选 ,获得了葡萄属植物的RAPD最优反应体系及扩增程序 .
An improved DNA extraction protocol with CTAB was developed for Vitis species. DNA was extracted successfully from the samples of 7 species and was suitable for RAPD analysis. Five important factors affecting RAPD were designed for screening and re screening the best RAPD reaction system. An economic and effective RAPD system was obtained which adapted to grape RAPD analysis in order to be used for large scale classification research of grape.
