内质网应激凋亡通路中的耐药节点 被引量:2

摘要 内质网应激(endoplasmic reticulum stress,ERS)介导的细胞凋亡是一种新的凋亡途径,ERS过长过强,可多途径诱导细胞凋亡。肿瘤化疗药物的作用机制是诱导肿瘤细胞发生凋亡,因此凋亡信号传导通路在肿瘤治疗中尤为关键。凋亡通路中任意环节缺陷都能导致肿瘤细胞对凋亡的耐受。该文就内质网中影响凋亡信号传导通路中发挥关键作用的一些重点耐药分子作一综述,包括P-gp分子、IAP家族、Bcl-2耐药家族中的关键蛋白以及内质网钙泵IP3R。 Endoplasmic reticulum stress (ERS) mediated apopto- sis is a new pathway of apoptosis. ERS is too long or too strong,which can induce cell apoptosis in many different ways. The mechanism of tumor chemotherapy drugs is to induce tumor cell apoptosis, so apoptotic signaling pathways in cancer therapy are particularly important. Any aspect of defects in these pathways can lead to the resistance of tumor cell to apoptosis. This review focuses on research achievements about key medicine resistance molecules in the endoplasmic reticulum stress mediated apoptosispathway, including the P-gp molecule, the key proteins in lAP family and Bcl-2 family as well as the endoplasmic reticulum cal- cium pump IP3R.
