摘要
目的:研究不同强度跑台训练对BALB/c小鼠气道muc5ac和aqp5表达的影响,探讨运动性支气管痉挛(EIB)发生的潜在机理。方法:清洁级雌性BALB/c小鼠48只,随机分为4组:对照组(C组)、小强度运动组(L组)、中等强度运动组(M组)和大强度运动组(H组)。L组、M组和H组小鼠分别给予不同强度的跑台训练:速度分别为4 m/min(58%VO2 max)、12 m/min(76%VO2 max)、17 m/min(84%VO2 max),坡度均为0°,每周运动6天,持续2周,每次训练前均做10min热身运动(速度1m/min,跑台坡度0°)。用实时定量荧光PCR法检测气道内muc5ac mRNA和aqp5 mRNA的表达,分别用免疫组化法和免疫印迹法检测muc5ac蛋白和aqp5蛋白在小鼠气道的表达,对气道muc5ac蛋白含量与aqp5蛋白含量进行Spearman等级相关分析。结果:运动各组小鼠气道内muc5ac mRNA及其蛋白表达均明显高于C组(P﹤0.05),而aqp5 mRNA及其蛋白表达均显著低于C组(P﹤0.05);并且气道muc5ac蛋白含量与aqp5蛋白含量呈负相关(r=-0.826,P﹤0.001)。结论:运动可能通过改变气道内muc5ac和aqp5的表达而使气道反应性升高,从而潜在地增加了EIB的发病风险。
Objective : The purpose of the paper is to observe the effect of the different intensity treadmill training on mucSac and aqp5 expression in airway in BALB/c mice so as to study the potential mechanism of exercise - induced bronchoconstriction (EIB). Methods : 48 female BALB / c mice ( Clean - level) were randomly divided into 4 groups : normal control group ( C group), low - intensity training group ( L group ), moderate - intensity training group ( M group) and high -intensity training group (H group). L, M and H group mice were given different intensity treadmill training respectively for2 weeks: speed: 4 m/ min (58% VO2 max), 12 m/ min (76% VO2 max), 17 m ! rain (84% VO2 max) ; incline: 0 o, 6 d/w. 10 min warm -up exercises were done (speed:lm/min, incline: 0 ~) before training. Expression of mucSac mRNA and aqp5 mRNA in airway were detected by real time quantitative fluorescence PCR, and expression of mucSac and aqp5 proteinum by immunohistochemical method and Western Blotting respectively. The relevant analysis of protein content of mucSac and aqp5 in airway was conducted with the Spearman rank correlation analysis. Results: the expressions of muc5ac mRNA and proteinum in L, M and H group were significantly higher than in C group ( P 〈 0.05 ), and the expressions of aqp5 mRNA and proteinum in L, M and H group were remarkably lower than in C group (P 〈0.05). The expressions of mucSac proteinum was negatively correlated to aqp5 (r = -0.826, P 〈 0. 001 ). Conclusion: Exercise could increase airway reactivity by altering the expression of mucSac and aqpS, thus potentially increasing the risk of EIB.
