摘要 目的 研究促吞噬肽 (tuftsin)和肿瘤坏死因子 (tumornecrosisfactor α,TNF α)的相互关系。方法 建立大鼠有脾和无脾动物模型 ,通过静脉注射TNF模拟病理状态下血中TNF浓度升高 ,利用反向高效液相色谱法检测血清中促吞噬肽的水平。结果 半脾切除组和全脾切除组术后 2周促吞噬肽水平下降 (P <0 .0 0 1) ,并以后者为著。注射TNF各组注射后d 3,d 5血清中促吞噬肽水平显著降低 (P <0 .0 0 1) ,并随时间推移呈下降趋势。结论 高浓度的TNF明显抑制促吞噬肽的产生 ,二者之间存在负反馈调节作用。首次提出了TNF -促吞噬肽轴的概念和二者临床应用的设想。证明了血清中促吞噬肽水平依赖于脾脏细胞的总量 ,保留一定数量的脾脏细胞对维持机体的免疫功能非常重要。 Objective To study the relationship between tumor necrosis factor α (TNF α) and tuftsin. Methods Models of animals with and without spleen were established in rats. The serum tuftsin levels in rats that had been injected with 2.5×104 IU TNF α 5 days before were determined with reverse phase high performance liquid chromatography. Results The serum tuftsin level significantly decreased in both the total splenectomy group and the semi splenectomy group 14 days after the operation (P<0.001). After the injection of TNF α, the tuftsin level was markedly decreased with time prolongation in all the groups (P<0.001). Conclusions High dosed recombinant TNF α can inhibit the production of tuftsin in rats. There is a negative feedback between TNF α and tuftsin. A new concept of TNF tuftsin axis is formulized for the first time in this study. The relationship between the two factors and the application of them in combination are also discussed. It is proved that retaining partial splenic tissue is very important for the body′s immunocompetence.
