摘要
目的:研究晚期糖基化产物(AGEs)对大鼠血管平滑肌细胞A7R5增殖的影响及相关机制。方法:用AGEs处理A7R5细胞,相同条件BSA处理为对照组,Western blotting检测RAGE蛋白表达的变化,MTT比色法测定细胞活性。结果:AGEs处理A7R5细胞后,AGEs受体RAGE及细胞增殖水平均明显升高,但与EGCG共孵育能显著抑制AGEs诱导的RAGE表达增加及细胞增殖水平。结论:EGCG能够抑制AGEs诱导的大鼠血管平滑肌细胞增殖水平,这可能与EGCG抑制AGEs受体RAGE的表达相关。
Objective To investigate the effect of advanced glyeation end product - induced proliferation of vascular smooth muscle cells (VSMCs) A7R5 and explore the mechanism. Methods After cultured VSMCs were treated with advanced glycation end products (AGEs) or BSA,RAGE expression was detected by Western blot; cell variability was quantified by MTr assay. Results After cultured VSMCs were treated with AGEs,the level of protein RAGE and the proliferation rate of VSMCs was up -regulated. However,the RAGE ex- press and the proliferation rate of VSMCs was decreased co - incubation with EGCG in AGEs - treated group. Conclusion EGCG can inhib- it AGE~ -induced proliferation of vascular smooth muscle cells (VSMCs). It may relate to the AGEs -induced RAGE low expression.
