鼻咽癌细胞株γ射线照射后端粒酶活性及各组分表达的研究 被引量:3

摘要 目的 探讨γ射线照射鼻咽癌细胞株HNE1后 ,端粒酶活性改变情况以及与端粒酶各组分基因表达的关系。方法 HNE1在γ射线照射前和照射后不同时间 ,利用RT-PCR方法检测端粒酶亚单位人端粒酶RNA (humantelomeraseRNA ,hTR)、人端粒酶相关蛋白 (humantelomerase -associated 1,TP1)和人端粒酶逆转录酶 (humantelomerasereversetranscriptase,hTERT)基因表达情况 ,以及用TelomerasePCR -ELISA方法检测端粒酶的活性。结果 HNE1在γ射线照射后端粒酶各组分中 ,hTR和TP1mRNA改变不明显 ,而hTERTmRNA逐渐降低 ,端粒酶的活性也逐渐降低。结论 HNE1经γ射线照射后 ,hTERTmRNA表达逐渐下降 ,端粒酶活性亦逐渐降低 ,hTERTmRNA改变与端粒酶活性改变呈一致性 ,提示hTERTmRNA的表达可能在调节端粒酶活性中起关键作用。
