摘要
目的 通过测定新疆地区静脉吸毒人群中HIV-1(1型人免疫缺陷病毒)感染者CD4细胞、血浆细胞因子(IL-2,INFγ,IL-4和IL-10)水平和末梢血单核细胞(PBMC)产生细胞因子的活性,了解该人群中HIV-1感染者细胞免疫学特征及其病程的关系。方法 将36例HIV-1感染者分为2组,第一组CD4细胞≥500/ul,第二组CD4细胞〈500/ul。
Objective To know cellular immunological features of HIV 1 positive individuals who are drug users in
Xinjiang area by investigating CD4 cell, the levels of cytokiries (IL-2, IFNγ, IL - 4, IL-10) in plasma and the eytokine
production of the peripheral blood mononuclear cells (PBMC). Methods Based on their CD4 cell count 36 cases of HIV
1 infected individuals were divided into two groups, the first group, CD4 cell≥500/μl and, the second group, CD4 cell<
500/μl. CD4 cell and the concentration of the cytokines were measured by flow cytometry and KLISA, respectively. Also,
the eytokine levels in plasma from 13 seronegative persons were measured as the control. In addition, IL 2 and IFNγ in the
cultural supernatant of PBMC- obtained from 6 HIV- 1 negative individuals and 6 HIV - 1 - positive ones were evaluated in vitro. Results (1) The level of plasma IL 2 in the infected individulas was lower than that of seronegative subjects, and there was a positive correlation between the level of IL-2 decreasing and disease progression (P<0.05); the levels of plas
ma IFNγ, IL - 4 and IL-10 were not significantly changes in the HIV 1-positive indeviduls; (2) To be compared with the control, the autonomous production of IL 2 by PBMC from positive people was lower (P<0.05), but PHA induced production of IL- 2 had no significant change (P>0.05); both of the autonomous and PHA induced production of IFNy by PBMC in positive group were decreased (P<0.05). Conclusion (1 ) The decreasing of level IL 2 in plasma of HIV 1-infected individuals among IDUs in Xinjiang area is positively related with the disease progression (P<0.05), it is suggested that the level of plasma IL - 2 be as a predictor of the disease progression; (2) It can be supposed that the decreasing of level of IL - 2 in (he plasma be positive conjunction with the expression active of IL-2 by PBMC; (3) The results, the level of plasma IFNy with a rising trend, IL 4 and IL - 10 without significantly increasing, show that these infected people are still in relatively early stage of the disease course.
