摘要
fel2-1被报道为一个在IQM2(At3g13600)基因中插有一个T-DNA的突变体,在白光和蓝光条件下呈现出长下胚轴的表型.文章报道了对该突变体进行分子鉴定的结果.首先利用PCR方法,验证了T-DNA在IQM2中的插入位点.因为该突变体的表型与蓝光受体隐花色素1(cryptocrome 1,CRY1)突变体的表型极为相似,所以继而对fel2-1中的CRY1基因进行了分子鉴定.结果表明,在该基因的起始密码ATG后第1 231 bp处也存在着一个T-DNA.因此,可以确定fel2-1是一个在IQM2和CRY1基因中都插有T-DNA的突变体,是研究IQM2和Cry1基因互作的良好材料.
We previously reported thatfel2-1 is a T-DNA insertion line of IQM2 (At3g13600) gene and showed a long-hypocotyl phenotype when grown under white- and blue-light condition. Here, we reported the results of molecular identification on this mutant. Through PCR analysis, the T-DNA insertion site in IQM2 was further confirmed as shown in the homepage of the mutant pool. Because phenotype offel2-1 was similar to that of blue light receptor cryptocrome 1 ( CRY1 ) mutants, molecular identification on CRY1 gene was then carried out. It has been shown that there is a T-DNA inserting in CRY1 gene at 1231 bp downstream of its start code ATG. Therefore, it is suggested that f el2-1 is a T-DNA insertion mutant in both IQM2 and CRY1 gene and is a good material to study the interaction of these genes.
