摘要
目的探讨不同缺血时间对大鼠海马和纹状体再灌注损伤标志物葡萄糖调节蛋白78(GRP78)表达的影响。方法将SD雄性大鼠48只,随机分为6组,每组8只,分别为对照组、假手术组、缺血30 min组、60 min组、90 min组和120 min组。除对照组外,其他各组采用大脑中动脉阻塞法制造脑缺血再灌注损伤模型。假手术组只穿线,不阻塞,实验组分别阻塞30 min、60 min、90 min和120 min后再灌注。分别于再灌注后12 h和24 h处死4只动物,取海马和纹状体备用。用RT-PCR检测各组海马和纹状体GRP78 mRNA的表达。结果缺血再灌注12 h和24 h后,不同缺血时间组动物海马和纹状体的GRP78 mRNA表达均明显增多,与对照组和假手术组比较有统计学差异(P<0.01),以再灌注12 h各组变化更明显。再灌注12 h时缺血90 min组升高最明显,而再灌注24 h时缺血120 min组升高最明显。结论缺血和再灌注损伤能上调海马和纹状体的GRP78 mRNA表达。
Objective To explore the effects of GRP78 mRNA expression in rat's hippocampus and striatum after cerebral ischemia injury. Methods Grade Ⅱ clean male SD rats, weighing 280-320 g, were randomly divided into 6 groups : control group, sham operation group, blocked 30 min ,60 min ,90 min and 120 min group,8 rats per group. The models of cerebral ischemia-reperfusion injury were replicated by middle cerebral artery occlusion in rats. Rats were blocked 30 min, 60 min ,90 min or 120 min respectively. 4 animals were sacrificed and rats' brains were taken for reserve after reperfusion 12 h and 24 h. RT-PCR was used to detected expression of GRP78 mRNA in the hippocampus and striatum on different time. Results GRP78 mRNA were significantly increased in all blocked groups, which compare with the control group and sham operation group were significantly difference ( P 〈 0. 01 ). All the blocked group compared with the positive control group was significant difference in the hippocampus ( P 〈 0. 05, P 〈 0. 01 ). Conclusion Increasing GRP78 mRNA expression of hippoeampus and striatum concerned after cerebral ischemia injury in rats.
