摘要
无血清培养是诱导细胞凋亡的常规方法 ,具有简便、快速的特点 .用光学显微镜、荧光显微镜及透射电镜观察了人胚肺二倍体成纤维细胞 (KMB1 7)在无血清培养过程中细胞凋亡的形态学发生过程 .结果 :无血清诱导后 1h ,光镜下可见致密单层的成纤维状细胞出现疏松状态 ;3h后 ,少数细胞(1%~ 5% )出现圆缩 ;6h后 ,成纤维细胞全部由长梭形收缩为圆形 ,体积缩小 .荧光及电镜下可将细胞凋亡从形态上分为两个阶段 ,第一阶段细胞出现典型的凋亡特征 ,细胞圆缩、出芽、核染色质凝缩、成块状或新月状分布于核膜边缘 ;第二阶段 ,在细胞凋亡晚期出现坏死症状 :电镜下整个细胞崩解成碎片 .结果表明 :无血清培养诱导的凋亡在凋亡的晚期由于营养过度缺乏易出现二次坏死现象 .首次报道用于生物制品生产的人胚肺二倍体细胞在无血清诱导下凋亡发生的形态学特征 .
The apoptotic morphogenesis of human embryonic lung diploidfibroblast KMB l7 cell line was observed during serum-free culture under lightmicroscope, fluoroscope and transmission electron microscopy. The results showedthat the loose gaps were developed in closed confluent monolayer cell an hour afterinduction by light microscope; the shuttle-like fibroblasts became round obviously 3 hours later apoptotic process was divided into two phases, the first one, the cellsshowed typically apoptotic characteristics: round and shrunk, budding, nuclearchromatin condensation into dump or crescent; the second one, necrosis was detectedin the late phase of apoptosis.
