摘要
目的检测基因IL-4 C590T和TNF-αG308A位点多态性与青海藏族结核病易感相关性。方法利用PCR-RFLP法对藏族结核患者(病例组)及健康对照者(对照组)各25例进行基因IL-4 C590T和TNF-αG308A多态性检测,比较分析各基因型与结核易感关系。结果对照组IL-4 T590C位点T/T纯合子、C/T杂合子、C/C纯合子基因型频率分别为56%、36%、8%,病例组分别为64%、32%、4%,两组间基因型分布差异无统计学意义(P>0.05);对照组C、T等位基因频率分别为0.74、0.26,病例组分别为0.80、0.20,C和T基因频率也无明显差异(P>0.05);对照组TNF-αG308A位点G/G纯合子、G/A杂合子、A/A纯合子基因型频率分别为64%、28%、8%,病例组分别为60%、28%、12%,AA基因型在对照组及病例组中,相对于GG和GA基因组表达较低,但是对照组和病例组间并没有明显差异(P>0.05);对照组G、A等位基因频率分别为0.78、0.22,病例组中分别是0.74、0.26,但两组的G、A等位基因频率分布差异无统计学意义(P>0.05)。结论 IL-4 C590T和TNF-αG308A基因多态性与藏族结核病易感性无关。
Objective To detect the relationship between polymorphism of gene IL -4 C590T,TNF -or G308A and the susceptibility of Qinghai tibetan tuberculosis. Methods The polymorphism had been analyzed by PCR - RFLP in 25 cases of tibetan tuberculosis patients and 25cases of responsible controls, and compared the relationship between different genetypes and tuberculosis. Results the genotypes of IL -4 T590C TY, CT and CC in control group was 56% ,36% and 8%, respectively;whereas in tuberculosis group was 64% ,32% and 4% , respectively. Compared with IL -4 T590C TF and CT, the CC genotypes was shown a lower expression in all of the groups, but there was no obvious difference between control group and tuberculosis group ( P 〉 0.05 ). The IL - 4 T590C gene frequency of C and T in control group was 0.74 and 0.26, respectively,in tuberculosis group was 0.80 and 0.20, respectively, there was no significant difference between the two groups ( P 〉 0.05 ). the genotypes of TNF - α G308A GG,GA and AA in control group was 64% ,28% and 8% ,respectively;whereas in tuberculosis group was 60% ,28% and 12% ,respectively. Compared with TNF-αG308A GG and GA,the AA genotypes were shown a lower expression in all of the groups, but there was no obvious difference between control group and tuberculosis group( P 〉 0.05 ). The TNF - c~ G308A gene frequency of G and A in control group was 0.780 and 0. 222, respec- tively,in tuberculosis group was 0.74 and 0.26, respectively, there was no significant difference between the two groups ( P 〉 0.05 ). In summery, there was no difference between tuberculosis morphism in the present study. Conclusion sis and the polymorphism of IL -4 C590T There are no relationship between and TNF-or G308A. and control group involved gene poly- the susceptibility of tibetan tuberculosis and the polymorphism of IL - 4 C590T and TNF -αt G308A.
