摘要
目的观察七氟醚对人红细胞抗氧化能力的影响,探讨其作用机制。方法健康成年人红细胞制成2%悬液,分为8组:单纯空气组(A组)、1%七氟醚组(S1组)、3%七氟醚组(S3组)、5%七氟醚组(S5组)、空气+过氧化氢(H2O2)组(A+H组)、1%七氟醚+H2O2组[(SI+H)组)]、3%七氟醚+H2O2组(S3+H组)、5%七氟醚+H2O2组[(S5+H)组)],各组通醚时间为30 min,孵育15 h后,加入反应浓度为200μmol/L H2O2,继续孵育3 h后检测溶血率、过氧化氢酶(CAT)及丙二醛(MDA)含量,所得数据用SPSS 13.0软件统计处理。结果 (S3+H)组溶血率(%)(17.384±1.976)比(A+H)组(10.848±0.274)和S3组(12.417±0.716)明显增加(P=0.007,P=0.027)。与A组(17.301±4.222)相比,S3组(5.431±0.531)和S5组(5.175±0.658)CAT含量(U/mgHB)明显降低(P=0.009,P=0.009)。(S1+H)组(4.319±0.235)、(S3+H)组(8.257±1.389)和(S5+H)组(9.156±0.742)CAT含量均较(A+H)组(13.689±2.003)明显降低(P<0.05)。而MDA含量(μmol/gHB)(S1+H)组(3.547±0.898)和(S3+H)组(4.474±0.653)均比(A+H)组(2.654±0.509)增高(P=0.031,P=0.021)。结论七氟醚可降低红细胞抗氧化能力,其机制与抑制过氧化氢酶活性有关。
Objective To observe the effect of Sevoflurane on the anti-oxidant capacity of human erythrocyte in vitro. Methods Human erythrocytes isolated from healthy volunteers were made to 2% suspension and divided into 8 group : group A (air), group S1 (1% Sevoflurane), group S3 (3% Sevoflurane), group S5 (5% Sevoflurane), group (A+H) (air and H202), group (SI+H) (1% Sevoflurane and H202), group (S3+H) (3% Sevoflurane and H202), group (S5+H) (5% Sevoflurane and H202). Each group was incubated for 15 hours after being ventilated with Sevoflurane for 30 min. H202 was added at a concentrarion of 200 μmol/L and incubated further for 3 hours. Then, the levels of catalase and MDA in erythrocytes as well as the haemolysis rate were determined. Data were analyzed by SPSS 13.0. Results The ery- throeyte haemolysis (%) in group (S3+H) (17.384±1.976) increased significantly compared with group (A+H) (10.848± 0.274) and group S3 (12.417±0.716) (P=0.007, P=0.027). Compared with group A (17.301±4.222), the levels of CAT (U/mgHB) in group S3 (5.431±0.531) and group S5 (5.175±0.658) decreased significantly (P = 0.009, P = 0.009). Compared with group (A+H) (13.689±2.003), the concentration of CAT in group (SI+H) (4.319±0.235)and (S3+H) (8.257±1.389) and (S5+H) (9.156±0.742) all significantly decreased (/9 〈 0.05). The levels of MDA (μmol/gHB) in group (S3+H) (4.474±0.653) more increased than in group (A+H) (2.654±0.509) and group (SI+H) (3.547±0.898) (P = 0.021, P = 0.031). Conclusion Sevoflurane inhibits the anti-oxidant capacity of human erythrocyte in vitro, which is involved in the attenuation of catalase activity in erythroeytes.
