摘要
目的:研究狼毒大戟(Euphorbia fischeriana Steud.)中巴豆烷(Tigliane)型二萜的抗肿瘤活性。方法:采用硅胶柱色谱、ODS、Sephadex LH-20分离化学成分,根据理化性质和波谱数据确定化合物结构,并对得到的化合物进行抗肿瘤活性研究。结果:从狼毒大戟中共分离鉴定了3个巴豆烷型二萜化合物,分别为:prostratin(1)、Langduin A(2)和13-O-Acetylphorbol(3),同时利用肝癌HepG-2细胞、胃癌SGC-7901细胞和乳腺癌MCF-7细胞进行抗肿瘤实验。结论:在MTT抗肿瘤实验中,化合物prostratin对肝癌和乳腺癌有显著的抗肿瘤活性、Langduin A对胃癌和乳腺癌有抗肿瘤活性、13-O-Acetylphorbol仅对乳腺癌有抗肿瘤活性。
Objective: To study the antitumor activity of Tigliane type diterpenes from Euphorbia fischeriana Steud.Methods: Separated and repeatedly purified used silica gel,ODS,Sephadex LH-20,HPLC column chromatographic method.The chemical structures of the isolated components were further elucidated by the physicochemical characters and spectroscopic methods.Results: Three compounds were isolated and elucidated as prostratin A,Langduin A as well as 13-O-Acetylphorbol.Conclusion: In vitro anti-tumor experiments it shows that the prostratin is a certain degree of anti-tumor activity of HepG-2 and MCF-7,and Langduin A has the antitumor activity to SGC-7901 and MCF-7,13-O-Acetylphorbol has the antitumor activity to MCF-7.
