摘要
目的 为了阐明表皮生长因了受体(EGFR)在喉癌组织中表达的形态学定位及其表达量与非癌变组织的差别。方 法 运用免疫组化S-P染色技术检测45例喉癌和21例声带息肉组织中EGFR的表达情况。对其中21例喉癌和癌旁正常喉 粘膜及10例声带息肉采用逆转录聚合酶链式反应(RT-PCR)进行EGFR-mRNA定量分析。结果 EGFR阳性的棕色颗粒主 要定位于细胞膜,呈阳性着色的癌细胞弥漫性分布在喉癌组织中;而在声带息肉组织中,EGFR阳性着色细胞则主要分布于基 底层。 EGFR-mRNA在喉癌组织(0.967±0.865)和癌旁组织(0.869±0.734)表达水平明显高于声带息肉(0.205±0.047)(P< 0.05)。结论 结果提示EGFR的过量表达和分布异常是喉癌的特有表现。EGFR-mRNA定量测定有可能成为喉癌早期诊断 的辅助手段。
Objective To elucidate the topogrophic and quantitative differences of the expression of epidermal growth factor receptor (EGFR) between laryngeal carcinoma and non-carcinoma tissue. Methods Immunohistochemical method (S-P staining) was used to examine the expression of EGFR in 45 cases of larygeal carcinoma and 21 cases of vocal cord Polyp, and for the quantitative determination, reverse transcri ption -polymerase chain reaction technique (RT-PCR) was applied in 21 cases of laryngeal careinoma and para-carcinoma hssue, as well as 10 cases of vocal cord poly. Results Those brown particles which were positive EGFR were mainly located in the cell membrane. In the samples of vocal cord polyp, the positive cells stained in EGFR were demonstrated in the hasal cell layer, while in the larygeal carcinoma samples, all the positive cells were diffusely localized. The expression levels of EGFR-mRNA in the larungeal carcinoma (0.967?.865) and paracarcinoma tissue (0.869?.734) were significantly higher than that in the vocal cord polyp (0.205?.047)(p<0.05). Conclusion The overexpression and abnormal distribution of EGFR are the characteristic presentations of laryngeal carcinoma. The determination of EGFR-mRNA may be helful in diagnosis of laryngeal carcinoma.
