摘要 目的探讨重组人生长激素(rhGH)对颅咽管瘤ADAMDEC1表达的影响。方法颅咽管瘤细胞经培养24 h后分为实验组和对照组,实验组分别加入浓度为10 ng/ml、100 ng/ml和1 000 ng/ml的rhGH,对照组不加rh-GH,采用RT-PCR检测颅咽管瘤细胞ADAMDEC1 mRNA表达水平,Western bloting检测ADAMDEC1蛋白在颅咽管瘤细胞中的表达变化。结果随着rhGH浓度的增加,ADAMDEC1 mRNA的表达和ADAMDEC1蛋白的表达均增加,rhHG1 000 ng/ml组均为最高。且颅咽管瘤细胞ADAMDEC1 mRNA目的基因/内参照的吸光度比值和各组积分光密度(IOD)值比较(去除β-actin),rhHG 1 000 ng/ml组均为最高。结论 rhGH可使颅咽管瘤细胞ADAMDEC1表达增加,从而促进颅咽管瘤细胞的生长,这可能为颅咽管瘤患者的rhGH替代治疗提供一个临床指导依据。 Objective To investigate the influence of recombinant human growth hormone (rhGH) on the ADAM- DEC1 expression in craniopharyngioma cells in human. Methods Cultured craniopharyngioma cells were divided into experiment group (adding rhGH) and control group (not adding rhGH) . The expression of mRNA and protein in ADAMDEC1 were detec- ted by RT - PCR and Western bloting respectively. Results With the increasing concentration of rhGH, ADAMDEC1 mRNA and protein expression were also increased~ ADAMDEC1 expression was the highest when the rhHG concentration was 1 000 ng/ml. And in craniopharyngioma cell, ADAMDEC1 mRNA target gene/Internal reference absorbance ratio within each group and inte- gral optical density (IOD) value (removal of 13 -actin) in Western bloting were the highest when rhHG concentration was 1 000 ng/ml group. Conclusion The rhGH can increase ADAMDEC1 expression thus promoting the growth of craniopharyngioma cells, and this may provide a clinical basis for guiding rhGH replacement therapy for craniopharyngioma.
