摘要 文章研究了不同浓度的SNP对粒毛盘菌YMU50胞外多酚(LTEP)积累的影响及LTEP纯化工艺。与对照组相比,0.05mmol/LSNP与0.10mmol/LSNP浓度处理下LTEP最大积累量(第8天)分别提高了6.06%和12.12%,而0.20mmol/LSNP浓度处理下,LTEP积累量低于对照组。研究表明,通过筛选得到..80~100目聚酰胺树脂对LTEP的静态吸附性能与解吸效果最好。利用80~100目聚酰胺树脂对LTEP进行纯:化,单因素试验结果表明,最优吸附条件为:上样质量浓度为2mg/mL,pH值为4,速率为1BV/h;最优解吸条件为:乙醇体积分数为20%,洗脱液流速为1BV/h,乙醇用量为500mL,洗脱液pH值为8.5。采用Box-Be—hnken组合设计优化解吸条件,得到最优解吸条件为:乙醇体积分数为18%,洗脱液流速为0.98BV/h,乙醇用量为599.99mL,洗脱液pH值为9。在最优吸附与解吸条件下得到的LTEP的纯度为75.45%。 The effect of SNP of different concentrations on extracellular polyphenol accumulation of Lachnum YMU50(LTEP) and the purification conditions of LTEP were investigated. The maximum accumulations of LTEP (the 8th day) with 0.05 mmol/L SNP and 0. 10 mmol/L SNP treatment in- creased by 6.06% and 12. 12%, respectively, compared with the control, whereas the maximum accu- mulations of LTEP were lower than that of the control with 0.20 mmol/L SNP treatment. Results showed that polyamide resin with 80-100 mesh provided the best static adsorption and desorption properties. LTEP was purified by using polyamide resin with 80-100 mesh. Single factor experiments results showed that the optimal adsorption conditions were as follows, sample concentration was 2 mg/mL, sample pH value was 4, and sample flow rate was 1 BV/h; the optimal dynamic desorption conditions were: ethanol concentration was 20%, desorption flow rate was 1 BV/h, the amount of eth- anol was 500 mL, and pH value was 8.5. A Box-Behnken design was made for further optimization of desorption conditions, and the results were: ethanol concentration was 18%,desorption flow rate was 0.98 BV/h, the amount of ethanol was 599.99 mL, and pH value was 9. The purity of LTEP reached 75.45 % under the optimal adsorption and desorption conditions.
