槲皮素对人前列腺PC3细胞株增殖和凋亡影响

Effects of quercetin on proliferation and apoptosis of PC3 cell line

目的:观察槲皮素对人前列腺癌PC3细胞株增殖和凋亡的影响。方法:采用CCK-8法染色检测不同浓度的槲皮素对人前列腺癌PC3细胞株细胞生长的影响,计算IC50;流式细胞仪AnnexinV/PI双染法检测PC3细胞的凋亡率;western-blot检测bcl-2与bax表达。结果:槲皮素抑制PC3细胞增长作用明显,且呈浓度依赖性,IC50为189.19μmol/L;AnnexinV/PI双染法检测显示,10μmol/L以上浓度的槲皮素可诱导PC3细胞的凋亡,降低bcl-2/bax比值,与阴性对照组比较,差异有统计学意义(P<0.05)。结论:槲皮素可抑制PC3细胞的增殖,诱导其凋亡,机制可能与下调bcl-2/bax比值有关。

Objective：To investigate the effects of quercetin on proliferation and apoptosis of PC3. Methods：Growth inhibition was measured by cell counting kit-8, the rate of apoptosis was detected by flow cytometry with Annexin-V/PI staining,and the express of bcl-2/bax was detected by western-blot. Results： Quercetin effectively inhibited the growth of PC3 in a concentration-dependent manner,IC50 was 189.19 μmol/L. Quercetin could induce the apoptosis of PC3 in a concentration over 10 μmol/L （P 〈 0.05）; compared with the negative control group, the express of bcl-2/bax was decreased.Conclusion：Quercetin can inhibit the proliferation and induce the apoptosis of PC3,the mechanisms may related with downregulating the ratio of bcl-2/bax .
