狼毒大戟对小鼠Lewis肺癌增殖及转移的影响 被引量:3

摘要 目的观察中药狼毒大戟提取液对Lewis小鼠移植瘤细胞增殖及转移的影响。方法收集对数生长期细胞接种至C57BL/6小鼠右侧腋窝下并随机分组,狼毒组腹腔给上述提取液,对照组给予等量生理盐水连续7 d,处死小鼠计算抑瘤率,荧光显微镜下观察Lewis细胞凋亡变化,免疫组化检测小鼠PCNA、TSP-1的表达。结果对照组平均瘤重为(0.3311±0.0184)g,狼毒组平均瘤重为(0.2209±0.06093)g;荧光显微镜下,狼毒组凋亡的Lewis细胞明显多于对照组;对照组与狼毒组PCNA平均光密度分别为1696.47±79.70、1364.47±108.52;TSP-1平均光密度值分别为307.94±65.66、428.90±57.92。结论狼毒大戟在诱导细胞凋亡的同时可以抑制肿瘤细胞的增殖与转移,可能是一种很有前景的药物。 Objective To explore the ettect of Euphorbta P'tschertana 5teuct on proliferation anti metastasts m Lewis carcinoma of mice. Methods Twelve mices were transplanted subcutaneously to the fight limb of 1 ~106 Lewis cells per mice. After transplanted,the mice were separaed into 2 groups:control and LDE group. The LDE group were given with LDE intraperitoneal injection; control group With same-volume of normal saline; the two groups of mice were killed after 7 days, then, tumor were taken off, and weighed firstly for mesureing inhibition rate; the change of Lewis cells of different groups were observed under fluorescence microscope with Annexin-V FITC/PI staining; Immunohitochemistry methods were used to analyse' expression of PCNA and TSP-I. Results The mean weight of the subcutaneous tumors of two groups was 0.3311±0.0184 g, 0.2209±0.06093 g, respectively; the number of apoptosis cells in LDE group was more than that in control group under the fluorescence microscope; In control group, mean density of PCNA was 1696.47±79.70, LDE group was 1364.47±108.52; the mean density of TSP-1 in two group was 307.94±65.66, 428.90±57.92, respectively. Conclusion LDE plays an important role in inhibiting proliferation and metastasis of Lewis carcinoma that may be a drug having great develop ment foreground.
