舒郁宁心汤对焦虑大鼠行为及血清神经肽和皮质酮水平的影响

The Effect of Shuyuningxin Decoction on the Behavior and Serum Levels of Neuropeptide Y and Corticosterone of Anxiety Rats

目的观察中药舒郁宁心汤对动物孤养加不确定性空瓶刺激焦虑大鼠模型的行为学变化以及血清神经肽(NPY)、皮质酮(CORT)含量的变化的影响。方法大鼠随机分为正常对照组、空瓶应激焦虑模型组、地西泮治疗组、舒郁宁心汤小剂量治疗组、舒郁宁心汤大剂量治疗组。采用不确定性空瓶应激刺激的方法建立大鼠焦虑的模型,空瓶刺激的第2周开始进行大鼠给药,连续用药7 d;然后观察大鼠空瓶刺激时探究、修饰和攻击行为的变化,以及血清神经肽、皮质酮含量的变化。结果模型组大鼠的攻击行为、探究行为明显增加,修饰行为明显减少,与对照组比差异有统计学意义(P<0.01)。舒郁宁心汤干预后大鼠的探究和攻击行为明显减少,修饰行为明显增加,与模型组比差异有统计学意义(P<0.05)。空瓶应激模型组大鼠血清NPY、CORT含量比正常对照组明显升高(P<0.05);大剂量舒郁宁心汤干预后大鼠血清NPY、CORT含量明显降低,与模型组比差异有统计学意义(P<0.05)。结论舒郁宁心汤具有抗焦虑作用,其作用机理与降低血清NPY、CORT的含量有关。

Objective： To investigate the effect of Shuyuningxin Decoction on the behavior and serum levels of neuropeptide Y（NPY）and corticosterone（CORT） in uncertainty empty bottle induced anxiety model.Method： 50 Sprague-Dawley rats were randomly divided into 5 groups： normal control group,anxiety model group,anxiety model with diazepam group,anxiety model with high/low dose group of Shuyuningxin decoction.Except normal group,the other groups were stimulated by empty bottle.After the rate stimulate by empty bottle for one week,the treatment group will treated for 7 days with different drug.The changes of exploratory,grooming and attack behaviors were observed and the content of serum levels of CORT and NPY was detected in each groups.Results： The scorn of exploratory,attack behaviors was significantly increased in model group,compared with normal control group there was significant difference.The scorn of grooming behaviors was significantly decreased in model group,compared with normal control group there was significant difference（P 0.01）.Shuyuningxin could significantly decrease the scorn of exploratory and attack behaviors,and increase grooming behaviors,compared with model group there was significant difference（P 0.05）.The serum levels of CORT and NPY was significantly increased in anxiety model group,compared with Normal control group there was significant difference（P 0.05）.High dosage Shuyuningxin could significantly decrease the level of NPY and CORT in the serum,compared with model group there was significant difference（P 0.05）.Conclusion： Shuyuningxin Decoction has anti anxiety effect;the mechanism maybe connects with decreased the serum levels of NPY and CORT.
