半仿生提取-高效液相色谱法测定芪白平肺胶囊中五味子醇甲的含量

Determination of Schisandrin in Qibaipingfei Capsules by semi-bionic extraction-HPLC

目的建立了高效液相色谱测定五味子醇甲含量的方法,同时考察半仿生提取(SBE)法与水提(WE)法两种提取方法对芪白平肺胶囊中五味子醇甲表征物质含量的影响。方法样品分别用WE和SBE法提取,经硅藻土净化,再用三氯甲烷提取,蒸干,甲醇定容,采用Welch materials(250mm×4.6mm,5μm)色谱柱分离,以甲醇-水(65:35)为流动相,流速1mL·min-1,检测波长230nm,柱温25℃,以标准曲线定量。结果实验结果表明,五味子醇甲在0.0228～0.2280mg·mL-1浓度范围内线性关系良好,相关系数为0.9999,平均加样回收率为99.31%,相对标准偏差(RSD,n=6)为0.29%。结论本方法简单、快速、灵敏,适用于五味子醇甲含量测定,且以五味子醇甲含量为指标时SBE法提取效率优于WE法提取。

OBJECTIVE To established a HPLC method for the determination of Schisandrn in Qibaipingfei Capsules. Meanwhile and Study on the influence of Schisandrin in Qibaipingfei capsules by WE method and SBE （semi-bionie extraction） method. METHODS The sample were extracted by WE method and SBE method,cleaned up by kieselguhr, then extracted by chloroform again, evaporated to dryness, metered volume by methanol, The separation was performed on a welch materials column （ 250mm×4.6mm,5μm）, methanol-water （ 70：30 ） as the mobile phase at a flow rate of 1mL · min^-1 and detector set at 230nm,the eolumn temperature was 25℃. RESULTS The results showed that there was good linear relationship between the mass concentration and the peak area of Schisandrin in the range of 0. 0228g · L^-1～0. 2280g · L^-1 with the correlation coefficient of 0. 9999（n =6） ,The average recovery was 99. 31% ,and the preeision（RSD） was 0. 28%. CONCLUSION This method is simple,accurate and suitable for the determination of Schisandrin. SBE method has higher efficiency of extraction than WE method with sehisandrin as index.
