用HPLC法测定积雪草苷涂膜剂中积雪草苷和羟基积雪草苷的含量 被引量:6

摘要 目的:建立用HPLC法同时测定积雪草苷涂膜剂中积雪草苷和羟基积雪草苷的含量。方法:采用Capcellpak C18色谱柱(250mm×4.6mm,5μm),流动相:甲醇-水(68∶32),流速:0.8ml/min,检测波长:204nm。结果:积雪草苷和羟基积雪草苷浓度在0.156~0.780mg/ml(r=0.999 3)和0.164~0.820mg/ml(r=0.999 5)范围内具有良好线性关系,精密度、稳定性和重复性良好。积雪草苷和羟基积雪草苷低、中、高浓度的加样回收率分别为(98.1±0.98)%、(99.4±1.01)%、(99.9±1.21)%(n=3)和(98.2±1.12)%、(99.0±1.03)%、(100.2±0.86)%(n=3)。3批样品中积雪草苷和羟基积雪草苷的含量分别为标示量的(101.1±1.21)%、(100.0±0.48)%、(101.1±0.57)%和(100.5±0.69)%、(100.0±1.07)%、(100.5±0.83)%(n=3)。结论:该方法准确、灵敏,重现性好,可用于积雪草苷涂膜剂的含量测定。 Objective: To establish a HPI.C method for the determination of the contents of asiaticoside and madecassoside in Asiaticoside film coating. Methods: Separation was performed on a Capcellpak C^s column (250 mmX 4.6 mm,5 /tin) with the mobile phase consisting of methanol-water (68 : 32) at a flow rate of 0. 8 ml/min. Detection wavelength was 204 nm. Results: Asiaticoside and madecassoside presented a good linearity within the range of 0. 156-0. 780 mg/ml (r=0. 999 3) and 0. 164-0. 820 mg/ml (r=O. 999 5) ,respectively. The method had fine qualities of good precision, stability and reproducibility. The recovery rates of low, medium and high concentrations of asiaticoside and madecassoside were (98.1±0.98)%、(99.4±1.01)%、(99.9±1.21)%(n=3)和(98.2±1.12)%、(99.0±1.03)%、(100.2±0.86)%(n=3),respectively. The contents of asiaticoside and madecassoside in the three batches of sample were (101.1±1.21)%、(100.0±0.48)%、(101.1±0.57)%和(100.5±0.69)%、(100.0±1.07)%、(100.5±0.83)%(n=3),respectively. Conclusion: It is an accurate, sensitive and reproducible method for the determination of the contents of asiaticoside and madecassoside in Asiaticoside film coating.
