摘要
蛋白质剪切是内含肽介导的蛋白翻译后过程。探讨了一个基于内含肽介导的蛋白质剪切技术用于蛋白质纯化的方法,利用内含肽介导的蛋白剪接原理将内含肽与目的蛋白连接,通过几丁质结合域与几丁质树脂结合进行亲和纯化。得到的目的蛋白无亲和标签,可降低其对蛋白质结构和活性的影响,优于以往的蛋白质亲和层析方法。
Protein splicing is a self-catalyzed process mediated by inteins. In this paper, a memoct oi protein purification was descried that used intein-mediated protein splicing technique. This system applied intein-mediated protein splicing technique. Target protein was fused to intein, and then the fusion protein was purified by affinity chromatography through chitin binding domain. The intein- based purification technology could obtain protein without affinity tag and eliminate its deficiency, which could retain the target proteins better than classical purified procedures.
