铸造钴铬合金含银抗菌涂层对L929细胞周期及凋亡的影响

The influence of cobalt chromium casting alloy and Ag containing coating on the cell cycle distribution and apoptosis of L929 cells

摘要 目的:检测钴铬合金含银抗菌涂层对L929细胞周期及凋亡的影响。方法:应用等离子喷涂技术制备含银抗菌涂层钴铬合金试件作为实验对象,制备浸提液用于培养L929细胞。利用流式细胞术测试细胞周期及凋亡并与常规铸造的钴铬合金试件进行比较。结果:浸提液培养48 h后,G2期细胞周期比例涂层组为(8.23±0.39)%,钴铬对照组为(8.70±0.46)%(P>0.05);细胞早期凋亡率涂银组为(2.10±0.07)%,钴铬对照组为(2.08±0.13)%(P>0.05)。结论:涂银钴铬合金与钴铬合金比较对L929细胞周期及凋亡无显著差异,细胞相容性良好。 Objective: To study the influence of cobalt chromium alloy and Ag coating on the cycle distribution and apoptosis of L929 cells. Methods: Ag antimicrobial coating was prepared by plasma spraying technique on cobalt chromium alloy and the samples were used in the experimental group. Casting cobalt chromium alloy samples without coating were used in the control group. The extracts of the samples of the two groups were prepared respectively. L929 cells were cultured in the extracts for 48 hours. Flow cytometry method was used to test the cycle distribution and apoptosis of L929 cells and the results were compared by statistic analysis. Results : The cell population of G2 stage in experimental and control group was ( 8.23 ± 0.39 ) % and ( 8.70± 0.46 ) % ( P 〉 0.05 ), the cell apoptosis rate was (2.10 ± 0.07 ) % and (2.08 ± 0.13 ) % ( P 〉 0.05 ), respectively. Conclusion : Cobalt chromium alloy and Ag coating does not affect the cycle distribution and apoptosis of L929 cells, indicating that Ag coating may have good cytocompatibility.
