摘要
目的基于毛细淋巴管与毛细血管孔径之间差别,设计并合成1种新型大分子淋巴特异性右旋糖酐与甲苯胺蓝偶合物染料示踪剂。方法以甲苯胺蓝为原料,右旋糖酐为载体制备大分子染料。兔颈部淋巴染色实验中,以甲苯胺蓝为阳性对照,对比2种染料相应颈部淋巴管、前哨淋巴结(SLN)染色时间,舌体扩散范围,并进行了病理组织学检查。结果制得平均分子粒径为200nm的新型大分子右旋糖酐和甲苯胺蓝偶合物淋巴染色示踪剂。新型大分子染料淋巴管平均染色时间(19.7min)大于阳性对照组(14.3min),SLN平均染色时间(>4周)大于对照组(<2d),舌体平均扩散范围(10.5mm)小于对照组(20.0mm)。结论新型大分子染料较常用活性染料甲苯胺蓝具有染色时间长,不易弥散,淋巴选择性高等特点。
Objective Based on the different diameters of the capillary lymphatic vessels and capillaries, a new macromolecular lymphatic tracer was designed and synthesized. Methods Using toluidine blue as starting material, dextran as carrier, the target macromolecular dextran-toluidine blue conjugates were obtained. With toluidine blue as positive control, the fading time and diffusion range of the new macro- molecular dextran-toluidine blue conjugates were evaluated in the animal experiments. Results the aver- age molecular size of the new macromolecular dextran-toluidine blue conjugates was 200 nm, and the average staining time was 19.7 minutes, more outweigh than the time of toluidine blue which was 14. 3 minutes. Its average diffusion range was 10. 5 mm, less than its positive control. Conclusion compared with the toluidine blue as a tracer, the macromolecular dextran-toluidine blue conjugates have obvious advantages such as longer staining time, less dispersion and can be taken up selectively by lymphatic system.
