金丝桃苷对高糖、氧化下内皮细胞凋亡及细胞色素C、Caspase-9和Caspase-3表达的影响 被引量:7

摘要 目的:研究金丝桃苷(hyperoside,HP)对高糖和氧化损伤导致的内皮细胞凋亡及细胞色素C、Caspase-9和Caspase-3表达的影响。方法:体外培养内皮细胞,分为正常细胞组、正常细胞+HP组、高糖损伤组、高糖损伤+HP组、H2O2损伤组和H2O2损伤+HP组,采用MTT法测定内皮细胞的活力,并采用RT-qPCR法、Western blot法测定CytC、Caspase-9、Caspase-3的mRNA及蛋白表达水平。结果:HP能够显著拮抗高糖和氧化导致的内皮细胞损伤(P<0.01),使CytC和Caspase-3的mRNA和蛋白表达极显著降低(P<0.01),Caspase-9的mRNA表达显著降低(P<0.05),蛋白表达极显著降低(P<0.01)。结论:HP通过抑制CytC、Caspase-9、Caspase-3的表达而拮抗高糖、氧化损伤导致的内皮细胞凋亡,对DR血管内皮细胞具有一定的保护作用。 OIIJECFIVE To study the effect of hyperoside(HP) on the apoptosis and expression of CytC, Caspase-9 and Caspase-3 in endothelial cells under high glucose and oxidative damage. METHODS The endothelial ceils were cultured in vitro, and divided into normal cells group, the normal cells + HP group, high glucose injury group, high glucose injury + HP group, H202 injury group and H202 injury + HP group, using MTT to assay the activity of endothelial cells, and using RT- qPCR method, Western blot to assay mRNA and protein expression level of of CytC, Caspase-9,Caspase-3. liF.SULTS HP could significantly antagonize the injury of endothelial cells due to high glucose and oxidation (P〈0. 01). HP could significant ly reduce the mRNA and protein expression of CytC and Caspase-3 of endothelial cells (P〈0. 01). HP could significantly re-duce 1he rnRNA expression of Caspase-9 (P〈0. 05) and the protein expression level of Caspase 9 (P〈0. 0l). CONCLUSION By inhibiting the expression of CytC,Caspase-9,Caspase-3, HP can antagonize apoptosis of endothelial cells due to high glucose or oxidative dan:age. HP has a protective effect on DR vascular endothelial cells.
