口疡清方对复发性口腔溃疡大鼠的治疗作用及其对白细胞介素-2、白细胞介素-4表达的研究 被引量:5

Research of Kou Yangqing prescription for treatment of recurrent oral ulcer rats and its effect on IL-2,IL-4 expression

摘要 目的:研究口疡清方对复发性口腔溃疡(recurrent oral ulcer,ROU)模型大鼠的疗效,并通过白细胞介素-2(IL-2)、白细胞介素-4(IL-4)的表达探讨该方治疗ROU的可能机制。方法:(1)用免疫法建立大鼠ROU模型,镜下观察口腔黏膜变化。(2)用流式细胞术(flow cytometry,FCM)检测各组外周血CD3+、CD4+、CD8+比例,与对照组比较。(3)用酶联免疫吸附测定(ELISA)各组IL-2、IL-4的表达。结果:模型组CD3+(56.4±8.8∕63.1±1.7)、CD4+(42.9±5.7∕48.5±4.0),CD8+(27.3±4.2∕30.2±1.0),IL-2(0.51±0.04∕0.72±0.20)、IL-4(0.45±0.05∕0.57±0.09)表达较正常组均有所下降,经口疡清方干预后相应表达有所上升,其中,中剂量组(66.4±6.8,48.2±4.9,33.8±11.8,0.67±0.08,0.45±0.05)和左旋咪唑组(64.5±6.3;47.6±5.5,29.2±3.9,0.61±0.09,0.48±0.08)上升明显,与模型组比较有统计学意义(P<0.05)。结论:口疡清方对ROU有治疗作用,其作用的机制可能与IL-2、IL-4的表达有关。 To study therapeutic effects of Kou Yangqing prescription for the recurrent oral ulcer model rats, and explore the possible mechanisms of the treatment ROU by IL-2 IL-4 expression. METHODS (1) Immune method was used to set ROU rats model, and the oral mucosal changes was observed by microscope; (2)CD3* , CD4± , CD8± were determined by flow cytometry and compared with the control group; (3)IL-2, IL-4 expression were determined by ELISA. RESULTS In the modelrats CD3± (56. 4 ± 8.8 / 63.1 ± 1.7) ,CD4± (42. 9 ± 5.7 / 48. 5 ± 4. 0) ,CD8± (27. 3 ± 4. 2 / 30. 2 ± 1.0)was declined, also by IL-2(0. 51 ± 0. 04 / 0. 72 ± 0. 20) ,IL-4(0. 45 ± 0. 05 / 0. 57 ± 0. 09) was declined, and increased after drug intervention, the middle dose group (66. 4 ± 6. 8,48. 2 ± 4. 9,33. 8 ± 11.8,0. 67 ± 0. 08,0. 45 ± 0. 05)and levamisole group (64. 5 ± 6. 3,47. 6 ± 5.5,29. 2 ± 3.9,0. 61 ± 0. 09,0. 48 ± 0. 08) increased significantly compared with model group (P〈0. 05), with statistical significance. CONCLUSION Kou Yangqing prescription has therapeutic effect for ROU, its mechanism may be related to the expression of IL-2 IL-4.
