摘要
目的:建立用酶联免疫(ELISA)法测定去氨加压素血药浓度的快速、简便方法,并研究其药动学。方法:用人去氨加压素(dDAVP)ELISA试剂盒对8例受试者给药后各时间段的血样进行血管加压素含量测定,用SPSS 18.0对检测平均值作药时曲线图并计算药动学相关参数。结果:ELISA法测定去氨加压素标准曲线回归方程为:Y=92.364 X+9.655 5,R2=0.998 1,平均回收率为100.96%;t1/2=(81.1±10.9)min;Cmax=(175.1±8.8)pg·mL-1;AUC0→200=(18 415±964)pg·h·mL-1;ClT=(52.8±2.6)mL·min-1。结论:ELISA法用于测定去氨加压素血药浓度及药动学研究的方法快速方便,结果稳定可靠。
To develop a simple and rapid method for determining the concentration of desmopressin and study its pharmacokinetices. METHODS Blood samples of 8 patients collected at different time after administration were determined using ELISA kit. The AUC and pharmacokinetics parameters were calcultated by SPSS 18.0 software.
