15-KETE对大鼠肺动脉平滑肌细胞Kv1.2、Kv3.4表达的影响

The Effect of 15-KETE on the Protein and mRNA Expression of Kv1.2,Kv3.4 in Rat Pulmonary Arterial Smooth Cells

摘要 目的从细胞分子水平研究电压门控型钾通道亚型Kv1.2、Kv3.4在15-酮基二十碳四烯酸(15-ketoeicosatetretraenoic acid,15-KETE)致大鼠肺动脉收缩过程中的作用。方法通过酶法分离、培养SD大鼠肺动脉平滑肌细胞(pulmonary arterial smooth cells,PASMCs),使用RT-PCR和Western blot技术观察15-KETE对大鼠PASMCs上Kv1.2、Kv3.4通道表达的影响。结果 (1)15-KETE下调大鼠PASMCs膜上Kv1.2通道mRNA及蛋白质的表达;(2)15-KETE下调大鼠PASMCs膜上Kv3.4通道mRNA及蛋白质的表达。结论 Kv1.2、Kv3.4通道参与由15-KETE引起的缺氧肺动脉收缩(hypoxic pulmonary vasoconstriction,HPV)。 Objective To observe the effect of Kv1.2、Kv3.4 channels in rat pulmonary arterial smooth cells on the process of pulmonary vasoconstriction induced by 15-KETE.Method Western blot and PCR were used to identify the effect of 15-KETE on expression of Kv1.2、Kv3.4 channels in cultured rat pulmonary arterial smooth muscle cells(PASMCs).Result(1) The expressions of Kv1.2 mRNA and protein in rat PASMCs were significantly down-regulated by 15-KETE.(2) The expressions of Kv3.4 mRNA and protein in rat PASMCs were significantly down-regulated by 15-KETE.Conclusion Kv1.2、Kv3.4 channels are involved in vasoconstriction induced by 15-KETE in rat PASMCs.
