提高香蕉胚性愈伤诱导成功率的可能途径

摘要 香蕉是我国重要的热带亚热带农作物。由于绝大多数食用香蕉是三倍体,不能通过授粉进行遗传改良,转基因技术就变得尤为重要。建立胚性悬浮细胞系是进行香蕉遗传转化的第1步。但是,建立香蕉胚性悬浮细胞系的成功率极低,直到现在多数实验室还不能自己建立香蕉胚性悬浮细胞系。然而,为什么香蕉胚性悬浮细胞系如此难以建立,在香蕉体细胞向胚性细胞转化的过程中,发生了哪些关键事件,如何提高香蕉胚性愈伤诱导的成功率,关于这些问题,目前还未见到报道。文中对近年来香蕉胚性悬浮细胞系建立方面的工作进行综述,针对香蕉胚性悬浮细胞系建立过程中发生的关键事件进行阐述,对香蕉胚性悬浮细胞系建立困难的原因进行讨论,并提出了若干提高香蕉胚性愈伤诱导成功率的可能途径。 Banana is a key crop across the humid subtropics. Genetic transformation is very important for banana varieties improvement be-cause of their parthenocarpy and female sterility. However, constructing an embryonic cell suspension (ECS) line, the first step for banana genetic transformation, is a very hard work. It had not been a routine procedure for many labs until now. Meanwhile, why it is so hard to get banana ECS, what is the mechanism underlying this phenomenon, what happened when banana somatic cell was transformed into embryonic cell, how to improve the success rate for getting banana ECS, had not been studied up to now. This review summaries our current knowledge on these questions, and more specifically, recent advances in the mechanisms that how somatic cells were reprogrammed to generate embryonic cells.
