摘要
研究了影响大麦花药离体培养反应的一些因素。结果表明 :供体穗子经 3℃预处理可以促进花药离体培养反应 ,迟钝基因型的最佳处理时间比敏感基因型的短 ,以 10~ 15d为宜。诱导培养基中添加 80 0mg/LCH能够较大幅度提高花药反应率和绿苗率 ,在含CH的培养基内添加 50 0mg/L脯氨酸或 10 0mg/L精胺 ,均能进一步提高迟钝基因型的反应花药率 ,特别是绿苗率。
Several factors influencing barley anther culture response were investigated. The results indicated: the culture response was able to be improved by 3℃ cool pretreatment of donor's spikes and the suitable pretreatment time for low responding genotypes was shorter than that for high responding ones. The optimal coolpretreatment time was 10~15 days for low responding genotypes. The freq.of responding anthers and regenerated green shoots were significantly increased by adding 800mg/L CH to induction medium and the freq.of responding anthers, especially,the freq.of regenerated green shoots could be further promoted by addition of 500mg/L proline or 100mg/L spermine to induction medium with CH.
