摘要 目的:探讨自噬在大鼠骨髓间质干细胞神经分化过程中作用。方法:实验分为:对照组、不同浓度自噬抑制剂3-甲基腺嘌呤(3-MA)组、不同浓度自噬激活剂雷帕霉素(rapamycin)组,比较各组诱导神经元分化前后神经标志蛋白微管相关蛋白质2(MAP-2)、Tau蛋白(Tau)、胶质纤维酸性蛋白(GFAP)的差异及特异性标记自噬的微管相关蛋白轻链3(LC3B)的表达变化。结果:大鼠MSCs可以诱导分化为神经元,其中以10μmol/L雷帕霉素组最佳,神经元标志蛋白MAP-2、Tau表达显著高于其他各组(P<0.05)。结论:适度提高自噬活性可增加大鼠MSCs诱导后神经分化效率。
