调节性树突状细胞在直接识别和间接识别反应中的作用

Effect of regulatory dendritic cells on corneal allograft rejection

摘要 探讨调节性树突状细胞(regulatory dendritic cell,rDC)在移植排斥反应中对T细胞直接识别和间接识别反应的作用。在TGF-β和IL-10的作用下,将小鼠骨髓来源的树突状细胞(DC)培养成rDC,比较rDC和成熟DC在LPS刺激下表型、细胞因子和对同种异体T细胞的刺激活性,检测调节性DC在对受者小鼠的脾脏T细胞直接识别和间接识别的作用。rDC表达共刺激分子明显低于成熟DC,分泌高水平的IL-10,对同种异基因T细胞有明显的抑制作用。在体外中明显抑制了受者小鼠脾脏T细胞的直接和间接识别的反应性增殖。rDC具有明显的异基因抑制作用,对同种异体T细胞的直接识别和间接识别反应性增殖有抑制作用。 To examine the effects of bone marrow derived regulatory dendritic cells (rDC) on both direct and indirect allore sponses after high-risk transplantation, donor derived dendritic cells were treated with TGF -β and 0IL- 10 and stimulated with lipopolysaccharide(LPS) to produce mature and regulatory DCs. Expression of costimulatory molecules and cytokines in rDC was analyzed, and the effects of rDC on both the direct and indirect pathway of alloreactive T cells were evaluated, rDC relative to mDCs showed a resistance to up-regulation of costimulatory molecules, as well as an impaired capacity to stimulate alloreac rive T cells in trivo, and demonstrated a remarkable reduction in the frequency of both directly and indirectly allosensitized T cells in recipients. These findings suggest that regulatory DCs demonstrate potent immunoregulatory properties in alloreactive T cells in vitro, and suppress both the direct and the indirect pathways of allosensitization.
