脂肪间充质干细胞结合透明质酸钠对兔骨性关节炎治疗的实验研究 被引量:4

摘要 [目的]探讨通过脂肪间充质干细胞结合透明质酸钠治疗兔膝关节骨性关节炎。[方法]通过制备兔膝关节骨性关节炎的经典动物模型及体外培养兔脂肪间充质干细胞。将制备完成的模型分为四组,每组10只:A组为对照组,不予处理;B组给予脂肪间充质干细胞复合纤维蛋白胶(FG)局部移植治疗;C组给予关节腔注射透明质酸钠治疗;D组给予脂肪间充质干细胞复合纤维蛋白胶(FG)加关节腔注射透明质酸钠治疗。治疗3周后处死实验动物检查治疗情况。[结果]通过对实验动物膝关节内蛋白多糖、MMP-13、Ⅱ型胶原的mRNA检测发现,脂肪间充质干细胞加透明质酸钠注射对早期膝关节骨性关节炎具有治疗效果。蛋白多糖、Ⅱ型胶原的表达均较对照组有所升高,MMP-13的表达较对照组下降。[结论]脂肪间充质干细胞加透明质酸钠注射对早期膝关节骨性关节炎具有治疗效果。 [ Objective] To investigate the effect of adipose mesenchymal stem cells combined with hyaluronie acid sodium in treatment of the rabbit knee osteoarthritis. [ Methods] The classical animal models of rabbit knee osteoarthritis was made and the rabbit adipose mesenchymal stem cells were cultured in vitro. Animals were divided into 4 groups with 10 animals in each. Group A was control group, withnot any processing. Group B received local transplantation treatment of adipose mesenchymal stem cells in fibrin glue (FG) . Group C was given intra - articular injection of hyaluronic acid sodium. Group D was given adi- pose mesenchymal stem cells in fibrin glue (FG) and intra - articular injection of hyaluronic acid sodium. After 3 weeks, all la- boratory animals were sacrificed and the treatment effects were analyzed.[ Results] By detecting the proteoglycans, MMP - 13, type Ⅱ collagen mRNA, therapeutic effect of adipose mesenchymal stem cells combined with hyaluronie acid sodium was found on early knee osteoarthritis. Proteoglycan, type Ⅱ collagen expression increased in experimental group compared with the control group. MMP - 13 expression decreased in experimental group compared with the control group. [ Conclusion] Adipose mesenchymal stem cells combined with hyaluronie aeid sodium has obvious effect on early knee osteoarthritis.
