低温胁迫下软枣猕猴桃叶片蛋白质双向电泳分析与质谱鉴定 被引量:9

Analysis and identification of proteins from leaves of Actinidia arguta under low temperature stress by two-dimensional electrophoresis and peptide mass fingerprinting

摘要 【目的】探明软枣猕猴桃在低温胁迫前后蛋白质表达的差异性。【方法】以‘魁绿’软枣猕猴桃扦插苗为材料,在4℃控温箱中低温处理1 d后取其叶片进行双向凝胶电泳及质谱鉴定,并与未处理前的对照组进行比对。【结果】低温胁迫前后蛋白质表达有明显的上调或下调现象。通过丰度计算,得到差异显著(P<0.05)的蛋白点共63个。运用MALDI-TOF质谱分析从中选择的24个蛋白点,成功鉴定出11种蛋白质,包括与光合作用相关的蛋白、与糖代谢相关的酶、与基因调控相关的蛋白、与信号转导相关的酶、与抗逆性相关的酶等。【结论】低温胁迫前后,软枣猕猴桃叶片蛋白质表达存在明显差异,低温影响到软枣猕猴桃内部各种复杂的生物学进程。 [Objective]The objective of the study was to elucidate the protein expression differences under low temperature stress in A ctirtidia arguta Planch. [Method] The 'Kuilti' A ctinidia arguta cuttings were treated with low temperature(4 ℃) for one day and performed proteomics analysis by 2-DE and peptide mass fingerprinting with the untreated control. [Result]The results indicated that there were obvious differences in protein expression levels between the low temperature-treated and the untreated control. Total 63 different proteins (P〈0.05)were detected with abundance calculation. 24 proteins were analysed by MALDI-TOF-MS and 11 proteins were function-known which included proteins involved in photosynthesis process, sugar metabolism, gene regulation, signal transduction, stress resistance and so on.[Conclusion]The protein expression levels existed obvious differences in Actinidia arguta cuttings, and low temperature influenced various complex biological process in A ctinidia arguta Planeh.
