苯酚硫酸一步法测定白芍提取物中总糖含量 被引量:7

Determination of Saccharide in Extracts of Pareonia Albiflota by One-step Phenol-Sulfuric Acid Method

摘要 目的:建立白芍提取物中中总糖含量的测定方法。方法:通过单因素试验确定实验的最大吸收波长,硫酸浓度,显色液用量,加热时间和显色时间。实验条件确定后,从加样回收率,重复性,线性和范围几方面进行方法学验证。同时,将优化后的方法与传统方法进行对比。结果:最佳实验条件为:硫酸与水的体积比为5∶1;显色液体积为5 mL;加热时间为50min;显色时间为15 min。在最佳实验条件下,葡萄糖标准溶液在0.019~0.189 mg线性关系良好,回归方程为Y=7.488 X-0.000 2(R2=0.999 5)。三个比例的加样回收率为102.00%,102.15%,95.86%,RSD分别为2.08%,1.51%,2.24%;6份样品的重复性RSD为2.4%。结论:该方法准确可靠,操作简单,也可用于其他中药提取物的质量控制。 Objective: The aim is to establish the determination method of saccharide in Pareonia albiflota extracts.Method: The volume ratio of sulfuric acid and water,the volume of color liquid,100 ℃ water bath time,and ice bath time were studied by single-factor experiment.Result: The optimum experimental conditions were as following: 5∶1 of the volume ratio of sulfuric acid and water,5 mL color liquid,100 ℃ water bath for 50 min,and ice bath for 15 min.In the optimum experimental conditions,the standard solutions of glucose possessed good linear with absorbance when its concentration was 0.019-0.189 mg.The linear equation was Y = 7.488X-0.000 2(R2= 0.999 5).The recovery ratio of three degrees was 102%,102.15%,95.86%,the RSD was 2.08%,1.51%,2.24%.The RSD of six samples was 2.4%.Conclusion: The method is simple,accurate and reliable.It can be used for the quality control of traditional Chinese medicine.
