摘要
目的:探讨熊果酸对人肝癌细胞株SMMC-7721的抑制作用。方法:将不同浓度的熊果酸分别与人肝癌细胞SMMC-7721共同培养24,72 h后,采用中性红染色法测定吸光度(A),评定熊果酸对肝癌细胞株的增殖抑制作用;流式细胞术测定其对肝癌细胞凋亡及细胞周期的影响。结果:不同浓度熊果酸对SMMC-7721细胞的增殖有明显的抑制作用(P<0.05或P<0.01),呈时间和剂量依赖关系;24,72 h的半数抑制浓度(IC50)为12.59,8.32 mg·L-1。流式细胞仪检测结果表明,熊果酸12.5 mg·L-1对肝癌SMMC-7721细胞凋亡率14.07%,凋亡率随药物浓度的增加而上升;SMMC-7721细胞阻滞S期,作用与药物剂量呈正相关。结论:熊果酸对肝癌细胞株SMMC-7721增殖有明显的抑制作用,抗肿瘤作用机制可能与诱导细胞凋亡及细胞周期阻滞于S期有关。
Objective: To discuss the inhibition mechanism of ursolic acid on inhibiting the growth of human carcinoma cell SMMC-7721.Method: Human hepatocellular carcinoma cell SMMC-7721 were co-cultured with different doses of ursolic acid for 24 h and 72 h,respectively.The absorbance value was evaluated by neutral red staining method,evaluation of ursolic acid on the proliferation of hepatoma cells;and the apoptosis was detected by flow cytometry.Result: Different concentrations of ursolic acid on the proliferation of SMMC-7721 cells showed obvious inhibition effect(P 0.05 or P 0.01) with a time and dose dependent manner.The half inhibitory concentration(IC 50) of ursolic acid on SMMC-7721 cell strain was 12.59 mg·L-1and 8.32 mg·L-1,respectively.The result indicated that the apoptosis rate was up to 14.07%,ursolic acid arrested SMMC-7721 the cell strain to S by flow cytometry with a dose-dependent manner.Conclusion: Ursolic acid on the proliferation of SMMC-7721 hepatoma cell line has the obvious inhibitory action,antitumor mechanism may be related to induction of cell apoptosis and cell cycle arrest in S phase.
