糖尿病对前列腺的影响及胰岛素样生长因子-1在前列腺的表达变化 被引量:1

摘要 目的 观察糖尿病对大鼠前列腺的影响,检测前列腺组织中胰岛素样生长因子-1(IGF-1)表达,探讨糖尿病对前列腺产生影响的病理机制.方法 将成年雄性SD大鼠35只随机分为两组,糖尿病模型组20只,正常对照组15只.禁食12h,糖尿病组予链脲佐菌素(STZ) 65 mg/kg进行单次腹腔注射,对照组腹腔注射等体积的枸橼酸缓冲液,两组均给予普通饲料喂养,提供充足自来水,均自由进食水.9周后,选模全部成功.各组大鼠切取完整前列腺,剔除筋膜和脂肪,立即称湿重.标本经苏木素-伊红(HE)染色后,光镜下观察两组大鼠前列腺结构,免疫组织化学方法检测各组前列腺组织IGF-1表达.结果 糖尿病组大鼠前列腺湿重降低,与对照组比较[(201.29±24.81) mg比(934.15 ±165.35) mg],差异有统计学意义(P<0.01).糖尿病组大鼠前列腺指数降低,与对照组比较(0.80±0.06比2.47±0.34),差异有统计学意义(P<0.01).光镜下可见糖尿病组大鼠前列腺上皮细胞发生萎缩,高度减低、变得扁平、皱襞减少,导管变得较空虚,导管内分泌物减少.糖尿病组前列腺上皮IGF-1的表达明显减弱,与对照组比较差异有统计学意义(5.33±1.84比3.00±1.92,P<0.01).结论 糖尿病导致前列腺重量减轻及重量指数减小,结构紊乱,可能影响其功能,对雄性生殖产生影响.糖尿病状态下前列腺组织上皮IGF-1表达减少. Objective To investigate the impact of diabetes on the prostate and the expression of insulin-like growth factor-1 (IGF-1) in the prostate tissue in order to understand the pathological mechanism of diabetes on the prostate.Methods Thirty-five male Sprague-Dawley rats (200-250 g) were randomly divided into two groups:normal control group (NC,n =15),and diabetes model group (DM,n =20).After fasting for 12 h,the rat diabetic models were induced through intraperitoneal injection of STZ (65 mg/kg).Isochoric citric acid buffer solution was given in control group.Two groups were given the ordinary animal feed and adequate water.Nine weeks later,diabetic models were successfully established.The whole prostate was removed.The prostates wet weight without fascia and fat was measured and all specimens were fixed immediately.Using haematoxylin-Iraq red (HE) dyes,morphological changes of the prostate tissues in the diabetic rats were observed under microscope.The IGF-1 expression in the prostates was detetected by using immunohistochemical staining.SPSS 13.0 software was used to processe the experimental date.Results The prostate wet weight in diabetic group was obviously slighter than that in control group [(201.29 ± 24.81) mg vs.(934.15 ± 165.35) mg,P < 0.01].The prostate index in diabetic group was obviously lower than that in control group (0.80 ±0.06 vs.2.47 ±0.34,P <0.01).Light microscopy revealed the pathological changes of the prostates in diabetic rats.The IGF-1 levels in the prostatic epithelia of diabetic group were significantly lower than in normal group group (5.33 ± 1.84 vs.3.00 ±1.92,P < 0.01).Conclusion Diabetes can lead to the reduced prostste weight,the decreased prostate index,structure disarrangement of the prostate,which may influence the male fertility.The decrease of the IGF-1 expression in the prostate tissue may be one of the factors inducing prostatic lesions in diabetes.
