摘要
目的:探讨p16基因与干扰素(IFN)-α-2b对黑色素瘤A375细胞生长的影响。方法 :将pDC312CMV-p16质粒转染进黑色素瘤A375细胞;检测细胞内源性和外源性p16基因mRNA表达差异;予转染组与非转染组A375细胞不同浓度IFN-α-2b,光镜下观察细胞形态变化;MTT检测IFN-α-2b对细胞生长的抑制作用。结果:未转染组无p16基因表达,转染组表达p16基因;转染组较非转染组细胞抑制率高,且差异有显著性(P<0.05)。光镜下,转染组及IFN-α-2b处理后A375细胞均呈凋亡状态。IFN-α-2b可抑制A375细胞增殖,抑制作用随浓度增加而增强(P<0.05);转染与给药存在交互效应(P<0.05)。结论:p16基因和IFN-α-2b均能抑制黑色素瘤A375细胞增殖,且存在交互作用。
Objective To explore the effects of p16 and interferon-α-2b (IFN-α-2b) on the growth of melanoma A375 cells. Methods The recombinant plasmid of pDC312CMV-p16 was transfected into melanoma A375 cells. The difference of mRNA expression between endogenous and exogenous p16 in melanoma A375 cells was detected. The transfected or non-transfected A375 cells were co-cultured with IFN-α-2b at different concentrations. The changes in cell morphology was microscopically observed. The action of IFN-α-2b in inhibition of the growth of A375 cells was determined by MTT. Results p16 was not expressed in the non-transfeeted melanoma A375 cells but was expressed in the transfected cells. The rate of inhibition was significantly higher in transfection group than in non-transfection group (P 〈 0.05 ). Apoptosis was microscopically found in the transfected and non-transfeeted ceils co-cultured with IFN-α-2b. IFN-α-2b inhibited the proliferation of A375 cells in a does-dependent manner (P 〈 0.05). A interaction effect was existed between p16 and IFN-α-2b (P 〈 0.05). Conclusions Both p16 and IFN-α- 2b can inhibit the proliferation of melanoma A375 cells with a interaction effect.
